1.Background
Androgenetic alopecia (AGA) is a condition causing scalp hair thinning, affecting 60–70% of adults globally. Developing effective treatments and understanding the underlying mechanisms are crucial. Previous studies have demonstrated that 650-nm red light can stimulate hair growth.
2.Objective
This study aimed to investigate the effects and mechanisms of 650-nm red light in treating AGA using an ex vivo hair follicle culture model.
3.Methods
Hair follicles were collected from AGA patients undergoing hair transplants.The follicles were cultured in Williams E medium and treated with or without 650-nm red light.Gene and protein expression were analyzed using real-time RT-PCR and immunofluorescence staining.RNA sequencing was conducted to identify gene signatures influenced by 650-nm red light treatment.
4.Results
Hair Follicle Proliferation:
650-nm red light significantly promoted the proliferation of human hair follicles in the cultured-tissue model.
Hair Cycle Regulation:
Treatment with 650-nm red light delayed the transition of hair follicles from the anagen (growth) phase to the catagen (regression) phase.
Gene Expression and Mechanisms:
RNA-seq analysis revealed that genes related to leukocyte transendothelial migration, metabolism, and adherens junctions were significantly involved in the stimulation of hair follicles.
5.Conclusion
The study demonstrated that 650-nm red light effectively promotes hair follicle growth and delays hair cycle regression in AGA. The transcriptomic analysis suggests that red light treatment may reverse hair follicle miniaturization, offering promising insights into AGA management.
