Apart from cancer, Alzheimer's disease (AD) and dementia represent one of the most pressing health challenges facing the Western world today. Numerous clinical trials have not shown any significant benefits from tested drugs in stabilizing or reversing the progressive cognitive decline experienced by dementia patients. Although the pathological hallmarks of AD, such as beta-amyloid plaques and tau tangles, are well known, there is still considerable debate regarding the genetic and lifestyle factors that may predispose individuals to developing dementia. Photobiomodulation (PBM) refers to the therapeutic use of red or near-infrared light to stimulate healing, alleviate pain and inflammation, and prevent tissue damage. In recent years, PBM has been explored for various brain disorders, often applied non-invasively by shining light on the head (transcranial PBM).

 

This review explores the mechanisms of action of transcranial PBM in the brain and summarizes studies that have used this technique to treat animal models of AD. The outcomes of a limited number of clinical trials using transcranial PBM to treat patients with AD and dementia are also discussed.Proposed chromophores for PBM can absorb different wavelengths of light. It is important to note that there is considerable overlap between the chromophores and that near-infrared light absorbed by structured water is likely to have a longer wavelength (>950 nm).
